Spermidine and fisetin: promising, and still early — on purpose
Two compounds we include with clear evidence-stage flags, because pretending they're settled would break our own rules. A look at autophagy, senolytics, and mixed human data.

Not every ingredient in a formula carries the same weight of evidence. Two of ours — spermidine and fisetin — are included precisely because their mechanisms are compelling and their human data is not yet conclusive. We label them that way rather than rounding up.
Spermidine — emerging
Spermidine is a polyamine found in foods like wheat germ, aged cheese, and soy. Mechanistically it is one of the best-characterized natural inducers of autophagy, the cell's self-cleaning process by which damaged proteins and organelles are broken down and recycled. Autophagy declines with age, and restoring it is a recurring theme across longevity biology.
The human outcomes, though, are genuinely mixed:
- A small three-month randomized trial in older adults at risk for dementia reported improved memory performance on a mnemonic task (Wirth et al., 2018).
- The larger, longer, more rigorously designed SmartAge trial in adults with subjective cognitive decline did not confirm a cognitive benefit over placebo (Schwarz et al., Alzheimer's Research & Therapy, 2019 — study protocol).
Strong mechanism, unsettled clinical outcome. That is the textbook definition of "emerging," and it is where we place it. It supports cellular renewal.
Fisetin — early
Fisetin is a flavonoid pigment found in strawberries, apples, and onions, and it belongs to a newer class of compounds called senolytics — agents that selectively clear senescent cells. Senescent cells stop dividing but refuse to die, accumulating with age and secreting inflammatory signals (the "SASP") that degrade surrounding tissue.
In a screen of ten flavonoids, fisetin was the most potent senolytic. Intermittent late-life dosing in mice reduced senescence markers across multiple tissues and extended median and maximum lifespan (Yousefzadeh et al., EBioMedicine, 2018). The preclinical case is genuinely exciting. Human trials are still small and early — so fisetin earns an early evidence badge, one rung below spermidine. It supports healthy aging and cellular balance.
Why cover them at all
Because a library that only ever discussed "strong human" compounds would be honest but useless — it would contain about three entries. The interesting frontier of aging biology lives exactly here, in compounds with excellent mechanisms and unfinished clinical stories.
The rule we hold ourselves to is simple: cover them, and never dress them up. An early badge is not an insult. It is information. See the full grading and the primary studies.
